My fiance and I went to Rome for two days. We walked everywhere! Ended up in Vatican City one afternoon and decided to go see the Sistine Chapel. We didn't research before we went and just assumed it would be in the giant church (St. Peter's basilica). When we entered it was beautiful everywhere. There was an option to go up to the top levels so we assumed that's where the painted ceiling would be and started going up the stairs... Turns out we climbed all the way to the very top of the tower, on the roof. Gorgeous views of the city, it was amazing and I would definitely recommend going. There is an elevator option for more money. It wasn't until we made it all the way back downstairs and our feet were killing us that we actually asked someone where to find the Sistine chapel and we made it over to the Vatican Museum just before closing.
